(and some not so big ones)
Toxicokinetic-toxicodynamic (TKTD) modelling is an exciting field of ecotoxicology that aims to understand and predict toxic effects, rather than just describing them. A lot of work has been done in this field, especially over the last two decades. However, many open (and in some cases fundamental) questions still remain to be solved.

About the Book
Toxicokinetic-toxicodynamic (TKTD) modelling has been my research topic (and passion) since 2002. Over the years, I realised that there were many things that I wanted (or needed) to know that were still unknown. Furthermore, I realised that answering these questions was, in many cases, not easy at all (especially for a theoretician with no access to a lab). Here these questions are bundled together to, hopefully, inspire a new generation of ecotoxicologists to address these fundamental scientific questions about the mechanistics underlying the response to toxicants (and some practical issues as well).
To fully appreciate these questions, you should have a firm grip on the concepts of TKTD modelling in general, and the concepts underlying GUTS and DEBtox in particular. These topics are only very briefly introduced in this book; much more thorough introductions are provided in my other e-books. This book is (for now) math-free. I guess this is not a book that you would want to read from front to back; it is probably more for browsing and selecting particular sections to read.
The questions are grouped in chapters. Each chapter starts with a short introduction. Each section within each chapter deals with an open question (or topic of research), and references are provided at the end of each section. Each section is rather short (I aim for a maximum of two pages, including a figure and references), so each discussion is rather superficial. The references given provide more details (and more references to consider).

About the Author
Mechanistic modelling is still an under-utilised tool in ecotoxicology and stress ecology. It is my passion to promote these models through (applied) scientific projects, providing software solutions, teaching in courses, presentations at conferences, and through writing e-books. The models that I specialise in are so-called toxicokinetic-toxicodynamic (TKTD) models; in particular, models for survival (the GUTS framework) and sublethal effects (based on Dynamic Energy Budget - DEB - theory). These models are based on a rigorous simplification of biological complexity, and have the power to explain (and predict) life-history traits of organisms (e.g., growth, reproduction and survival) over time. Especially when considering exposure to stressors, such dynamic models are indispensable to make sense of the data.
I hold an MSc and PhD in Biology, and have been a professional modeller since 1992, working almost exclusively on DEB- and GUTS-based TKTD modelling since 2002. Currently (as of 2015), I run my own private company 'DEBtox Research', offering my expertise to clients from industry, contract laboratories, research institutes, and universities.

We pay 80% royalties on purchases of $7.99 or more, and 80% royalties minus a 50 cent flat fee on purchases between $0.99 and $7.98. You earn $8 on a $10 sale, and $16 on a $20 sale. So, if we sell 5000 non-refunded copies of your book for $20, you'll earn $80,000.
(Yes, some authors have already earned much more than that on Leanpub.)
In fact, authors have earned over $15 million writing, publishing and selling on Leanpub.
